1160 Selective clearing of senescent dermal fibroblasts attenuates ultraviolet radiation-induced photoaging in mice

Photoaging mainly occurs due to ultraviolet (UV) radiation, and is accompanied by increased secretion of matrix metalloproteinases (MMPs) degradation collagen. UV radiation induces cell senescence in the skin; however, role senescent cells photoaging remains unclear. Therefore, elucidate photoaging, we evaluated effect senolytics a mouse model investigated underlying mechanism their antiaging effect. In both UV-induced human dermal fibroblasts model, ABT-263 ABT-737 showed senolytic effects on fibroblasts. Moreover, found that several senescence-associated secretory phenotype factors, such as IL-6, CCL5, CCL7, CXCL12, SCF, induced MMP-1 expression fibroblasts, which decreased after treatment with ABT-737in vivo vitro. Both drugs attenuated induction MMPs decrease collagen density model. Our data suggest targeting may reduce skin aging phenotypes thus agents are promising options for photoaging.
